1. Introduction

Ciprofloxacin is a broad-spectrum fluoroquinolone antibiotic belonging to the fluoroquinolone class and widely used in the antibacterial therapy (). Its main mechanism of action involves inhibiting bacterial DNA gyrase and topoisomerase IV, thereby disrupting the bacterial DNA-modifying enzyme, which is essential for bacterial DNA repair, transcription, recombination, and repair (). The mechanism of its action is dependent on binding the bacterial DNA gyrase and topoisomerase IV to the DNA. The primary mechanism of action involves binding to these sites, resulting in the inhibition of bacterial DNA repair, transcription, recombination, and repair ().

Ciprofloxacin was developed as a broad-spectrum fluoroquinolone antibiotic, which has been extensively studied in human and animal models, including rats, dogs, and pigs (). In mice, administration of ciprofloxacin has been shown to reduce inflammatory cytokine levels and improve neutrophil chemotaxis (). However, the effects of ciprofloxacin on other organs, including heart and lung, remain limited, which may be due to its poor oral bioavailability (). In contrast, the effect of ciprofloxacin on human neutrophils is generally well-controlled, with few adverse effects, and ciprofloxacin is widely used to treat bacterial infections in humans. In animal models, the anti-inflammatory effect of ciprofloxacin has been well-documented, including enhanced neutrophil migration, increased IL-1β production, and reduced IL-6 release, and ciprofloxacin has been used to treat various bacterial infections in humans.

In this study, we examined the effects of ciprofloxacin on renal function and liver function in mice and rats using mice with and without bacterial infection. The data from the current study show that ciprofloxacin reduced renal function and liver function, and its effect on these parameters was similar to that of the control mice. This suggests that ciprofloxacin may have therapeutic potential in human patients with bacterial infections, and further evaluation of its clinical applications is needed.

2. Materials and Methods

2.1. Animals and Drug

BALB/c mice and male rats were purchased from the National Laboratory Animal Care Center (NHLAC) of China. The male mice (n=12 in each group) were purchased from the National Laboratory Animal Care Center (NHLAC) and were housed in polypropylene cages in polypropylene cages at 23±2°C. The rats (n=12 in each group) were maintained in polypropylene cages at 23±2°C with free access to a12h light/dark cycle. The animals were fed food and water ad libitum. The mice were acclimatized to the experimental conditions at an interval of at least 5 days. The mice were anesthetized with pentobarbital (1 mg/kg) and given orally a single intramuscular (i.m.) injection of 0.05 mg/kg piperacillin (Sigma-Aldrich, St. Louis, MO, United States) every other day for a total of 10 days. The kidneys and bladder were harvested for histopathological evaluation. The mice were euthanized using an overdose of anesthetics (anesthesia was induced by inhalation of chloral hydrate) after the last injection of 0.1 mg/kg piperacillin (Sigma-Aldrich, St. Louis, MO, United States) into the kidneys. The bladder, kidneys, and other organs were immediately dissected out for histopathological evaluation.

2.2. Bacterial Infection

The mice were anesthetized with pentobarbital (1 mg/kg) and intramuscular (i.m.) injection of 0.05 mg/kg piperacillin (Sigma-Aldrich, St.

All animal procedures were conducted in accordance with the guidelines of the Institutional Animal Care and Use Committee of Nanchang University. The animals were randomly allocated into four groups and given the following experimental groups: Group I (n=10), ciprofloxacin (0.5 mg/kg, piperacillin;+0.05 mg/kg, piperacillin;0.
